Abstract

Tuning the physicochemical properties of nanoparticles to engineer the nanoparticle surface and/or payload is the first step towards engineering precision nanomedicines for therapeutic applications. Despite the clinical success of nanoparticles, particularly lipid nanoparticles (LNPs), there are still key challenges which are creating a bottleneck for translation to the clinic. A key barrier to rational design is the ability to relate nanoparticle structure to performance. An additional complexity can also be the existence of populations of particles within a single formulation.

Our studies have shown that several factors can impact the structure of nanoparticles including formulation methods, composition, payload and surface labelling. By combining small angle scattering (SAXS / SANS) to measure the average structure of nanoparticle formulations together with single nanoparticle analysis (fluorescence / Raman (SPARTAÒ)) we are aiming to map structure and function relationships in nanoparticle formulations to advance our understanding of existing pharmaceuticals and develop the next generation of nanomedicines.

Bio

Hanna Barriga is a Research Specialist and Project Leader in the Department of Medical Biophysics and Biochemistry at Karolinska Institutet in the group of Prof. Molly Stevens. She received her PhD in Chemical Biology of Health and Disease from Imperial College London from the Department of Chemistry and moved to Sweden in 2016 with a Marie Skłodowska-Curie fellowship. Recently she was awarded the VR starting grant and ERC starting grant. Her research centers around using nanotechnology to solve biological challenges. This multidisciplinary approach is focused on advancing our understanding of the biophysics of lipids and nanoparticles in biological processes to accelerate the development of the next generation of engineered nanomedicines.
